Separation of epigallocatechin-3-gallate from crude tea polyphenols by using Cellulose diacetate graft

摘要: This study demonstrates a new Cellulose diacetate graft -cyclodextrin (CDA- -CD) copolymer asymmetric membrane prepared by a phase inversion technique for the separation of (-)-epigallocatechin-3-gallate (EGCG) from other polyphenols in crude tea. The graft copolymer, CDA- -CD, was synthesized by prepolymerization of cellulose diacetate (CDA) and 1,6-hexamethylene-diisocyanate (HDI), which was then grafted with -cyclodextrin ( -CD). Surface and cross-section morphologies of the CDA- -CD membranes were analyzed by using scanning electron microscopy (SEM). Fourier transform infrared spectroscopy (FT-IR) indicated that the -CD was grafted onto the CDA by chemical bonding. The influences of the HDI/CDA mass ratio and the catalyst mass fraction on the -CD graft yield were investigated. The optimum conditions of a HDI/CDA mass ratio of 0.35 g·g and a catalyst mass fraction of 0.18 wt-% produced a -CD graft yield of 26.51 wt-%. The effects of the -CD graft yield and the concentration of the polymer cast solution on the separation of EGCG were also investigated. Under optimum conditions of a -CD graft yield of 24.21 wt-% and a polymer concentration of 13 wt-%, the purity of EGCG increased from 26.51 to 86.91 wt-%.

摘要:

The application and development of ‘green’ preventive technologies in tea plantations is an important means of ensuring tea quality and ecological safety. Ecological, agronomic and biological controls are the main preventive measures used in Guizhou Province. This paper summarizes the ‘green’ preventive technologies being applied in Guizhou tea plantations, including the use of plant defense inducers to regulate tea plant responses to pathogens, natural enemies to control pest species causing damage to shoots and Trifolium repens to control the main weed species. In addition, it summarizes the integrated ‘green’ preventive technologies being used in Guizhou and provides a foundation for the ecological maintenance of tea plantations.

TRANSCRIPTOME ANALYSIS REVEALS METABOLITE VARIATION AND FLAVONOID REGULATORY NETWORKS IN FRESH SHOOTS OF TEA

Metabolites, especially secondary metabolites, are very important in the adaption of tea plants and the quality of tea products. Here, we focus on the seasonal variation in metabolites of fresh tea shoots and their regulatory mechanism at the transcriptional level. The metabolic profiles of fresh tea shoots of 10 tea accessions collected in spring, summer, and autumn were analyzed using ultra-performance liquid chromatography coupled with quadrupole-obitrap mass spectrometry. We focused on the metabolites and key genes in the phenylpropanoid/flavonoid pathway integrated with transcriptome analysis. Multivariate statistical analysis indicates that metabolites were distinctly different with seasonal alternation. Flavonoids, amino acids, organic acids and alkaloids were the predominant metabolites. Levels of most key genes and downstream compounds in the flavonoid pathway were lowest in spring but the catechin quality index was highest in spring. The regulatory pathway was explored by constructing a metabolite correlation network and a weighted gene co-expression network.

Optimization of methyl orange removal from aqueous solution by response surface methodology using spent tea

摘要: The effective disposal of redundant tea waste is crucial to environmental protection and comprehensive utilization of trash resources. In this work, the removal of methyl orange (MO) from aqueous solution using spent tea leaves as the sorbent was investigated in a batch experiment. First, the effects of various parameters such as temperature, adsorption time, dose of spent tea leaves, and initial concentration of MO were investigated. Then, the response surface methodology (RSM), based on Box–Behnken design, was employed to obtain the optimum adsorption conditions. The optimal conditions could be obtained at an initial concentration of MO of 9.75 mg·L , temperature of 35.3°C, contact time of 63.8 min, and an adsorbent dosage 3.90 g·L . Under the optimized conditions, the maximal removal of MO was 58.2%. The results indicate that spent tea leaves could be used as an effective and economical adsorbent in the removal of MO from aqueous solution.

Alcohol and tea consumption in relation to the risk of nasopharyngeal carcinoma in Guangdong, China

摘要: To investigate whether alcohol and tea consumption has an etiological association with nasopharyngeal carcinoma (NPC) in a high-incident population, a large scale case-control study was conducted. The study included 2846 individuals in Guangdong Province, China, with 1387 newly diagnosed cases of NPC and 1459 frequency-matched controls. Exposure histories of alcohol and tea consumption were obtained via personal interviews. Information regarding socio-demographic characteristics (age, sex, education, dialect and household type), family history of NPC, Epstein-Barr virus (EBV) infection, dietary habits and other potential confounding factors was also studied. An analysis was performed using unconditional logistic regression to calculate odds ratios (OR) and 95% confidence intervals (CI). The risk of NPC was found to be associated with habitual alcohol consumption and tea consumption. Tea consumption has been associated with a decreased occurrence of NPC (OR= 0.62), while consumption of alcohol was associated with a complex effect. Specifically, moderate consumption of alcohol was associated with decreased risk of NPC, while overuse, especially strong distillate spirits, appeared to be a risk factor.

The essential role of immunoglobulin G (IgG) in immune system regulation and combatting infectious diseases cannot be fully recognized without an understanding of the changes in its N-glycans attached to the asparagine 297 of the Fc domain that occur under such circumstances. These glycans impact the antibody stability, half-life, secretion, immunogenicity, and effector functions. Therefore, in this study, we analyzed and compared the total IgG glycome—at the level of individual glycan structures and derived glycosylation traits (sialylation, galactosylation, fucosylation, and bisecting N-acetylglucosamine (GlcNAc))—of 64 patients with influenza, 77 patients with coronavirus disease 2019 (COVID-19), and 56 healthy controls. Our study revealed a significant decrease in IgG galactosylation, sialylation, and bisecting GlcNAc (where the latter shows the most significant decrease) in deceased COVID-19 patients, whereas IgG fucosylation was increased. On the other hand, IgG galactosylation remained stable in influenza patients and COVID-19 survivors. IgG glycosylation in influenza patients was more time-dependent: In the first seven days of the disease, sialylation increased and fucosylation and bisecting GlcNAc decreased; in the next 21 days, sialylation decreased and fucosylation increased (while bisecting GlcNAc remained stable). The similarity of IgG glycosylation changes in COVID-19 survivors and influenza patients may be the consequence of an adequate immune response to enveloped viruses, while the observed changes in deceased COVID-19 patients may indicate its deviation.
